The Respiratory Research group at the University of Dundee specialises in studies into the immunology and microbiology of respiratory diseases such as bronchiectasis, COPD and pneumonia. You will be expected to provide high quality technical assistance to Professor Chalmers and the other members of the research team. As a research technician you will play an important role in studies with direct benefit to patients such as clinical trials and patient cohort studies working alongside other research technicians within the group and overseeing the timely completion of research projects in partnership with the principal investigator, postdoctoral researchers and other research staff.

Core laboratory tasks may include processing biological samples, culture of airway epithelial cells at air liquid interface, microscopy, ELISA, and preparation of cells and biological samples for downstream processing such as flow cytometry and proteomics. You will support the principal investigator, post- doctoral researchers and PhD students as part of a multidisciplinary team. You will work collaboratively with team members to support multiple ongoing research studies where appropriate. You will work with external collaborators and internal department such as the Clinical Research Centre and core facilities to ensure the timely completion of research studies.

We are recruiting for a Grade 5 Research Technician

An opportunity has arisen for a research technician to join us within the Jackson Lab at the University of Dundee. This group will be exploring the role of Apolipoprotein E in Alzheimers Disease with an aim to target this important protein to prevent against Alzheimers Diseases.

You will be expected to provide high quality technical assistance to Dr Rosie Jackson and the other members of the research team. Core tasks will include culture of induced pluripotent stem cells derived astrocytes, HEK 293 cells, and primary astrocytes, and multiple molecular techniques including Maxiprep, ELISA, as well as in house assays of inflammation and protein thermal melt.

You will work with the principal investigator and overseas collaborators, performing laboratory research to a high standard and organising study data. You will work collaboratively with other team members including Postdoctoral fellows and PhD students to develop and validate protocols for sample collection and support curation of samples in long-term storage whilst supporting recruitment and sample collection to newly set-up clinical studies. You will be expected to work with the principal investigator to deliver research studies to a high standard in a timely manner.

We investigate mechanisms underpinning Bacillus subtilis biofilm formation and conduct interdisciplinary studies combining classical molecular genetics, microbiology, and soft matter physics.

We aim to uncover key regulatory mechanisms underpinning Bacillus subtilis biofilm matrix remodelling, function, resilience, and competitiveness, that direct the ability of this organism to occupy a wide range of environmental niches. The successful applicants will exploit recent findings to uncover the molecular processes driving adaptation of the biofilm matrix and the impact for the function and success of the bacteria in different environments.

We are recruiting for an exceptional individual to join as a Postdoctoral Researcher in the laboratory of Dr Arpan Mehta. This is a fixed-term appointment for 18 months, with potential extension at the end of the term.

We are recruiting for a driven individual to join the laboratory of Dr Arpan Mehta, a clinician-scientist, with expertise in cell biology and/or gene-editing to conduct an innovative pooled CRISPR screen in human motor neurons derived from induced pluripotent stem cells. The overarching goal of the lab is to undertake fundamental research to understand the molecular basis of amyotrophic lateral sclerosis (ALS), a form of motor neuron disease (MND), through open and interdisciplinary collaborations within and outwith of our unit.

The successful applicant will undertake a project as part of the MND Accelerator (MNDAcc) initiative funded by the Medical Research Council and National Institute for Health and Care Research to make the translation of discoveries into therapeutics easier for motor neuron disease and frontotemporal dementia. The goal is to make robust discoveries and share data openly in the space of 18 months.

Your priorities will include:

• Build C9ORF72-iCas9 iPS cell line and perform quality control followed by differentiation to motor neurons and microglia using published protocols.
• Optimise and perform a kinome-wide CRSIPR/Cas9 knock out-screen and sgRNA enrichment analysis.
• Validate the top candidate hits and create a CROP-seq library in partnership with UK Dementia Research Institute (UK DRI) single-cell facility.

We are recruiting highly motivated postdoctoral researchers to participate in a new academic-industrial global initiative supported by the Michael J Fox Foundation for Parkinson's Research. The project aims to: (1) Explore and validate new approaches to target LRRK2; (2) Perform systematic mechanistic, structural and safety analyses to benchmark new LRRK2 modifiers that emerge from this effort and (3) identify clinically relevant biomarkers for LRRK2-driven Parkinson's disease that will facilitate the selection of patients who respond to LRRK2 inhibitor therapy. We aim to galvanise research and accelerate the development of Parkinson's disease therapeutics. These positions enable further training in state-of-the-art methods through the course of the project, and involve wider collaboration with Protein Phosphorylation and Ubiquitylation Unit members, other academic Institutions worldwide and industrial partners working in this area.

We are looking for an exceptional Postdoctoral Research Fellow with strong Cell and Molecular Biology experience to join the Sapkota Lab to work on an exciting & innovative industry-academia collaborative project aimed at targeting membrane receptor and extracellular proteins for degradation through induced-proximity platforms. The field of targeted protein degradation, for example through PROTACs and molecular glue degraders, is emerging as an exciting new modality for research and therapeutics. The fellow will be based in the Sapkota lab (MRC-PPU, School of Life Sciences) and will work in close collaboration with teams at a leading international pharmaceutical company, which is funding this exciting interdisciplinary project. The Sapkota lab has developed several induced-proximity platforms for targeted protein degradation, phosphorylation, and dephosphorylation of intracellular proteins. This project seeks innovative approaches to push the boundaries to achieve proximity-induced degradation of cell membrane and extracellular proteins. This is a unique opportunity to gain experience in world-leading, collaborative and cutting-edge research in one of the hottest areas of modern translational chemical biology and drug discovery. We are recruiting for an exceptional individual to join us as a Postdoctoral Research Assistant within our Antigen Discovery Group, to support laboratory-based research in T cell antigen discovery. This is a fixed-term appointment for 3 years.

We are looking for a highly motivated Research Assistant with enthusiasm for T cell immunology and experience in sample preparation, analysis and interpretation of liquid chromatography mass spectrometry (LC-MS)-generated datasets. You will join the Antigen Discovery Group which aims to decipher the molecular architecture of the Human Leukocyte Antigen (HLA) processing and presentation machinery and study the effects of its function on T cell recognition in several disease models, including cancer. Processing samples of a variety of origins using standard lysis and immunoprecipitation, chromatography and mass spectrometry workflows, and data analysis will be an integral part of the position.

The position is central to the Antigen Discovery Laboratory and will support cutting-edge research ongoing in the wider institute. You will have the opportunity to formulate your own research projects and focus. You will have a background in Biology/Biochemistry (or a related discipline) with considerable bench science experience, and ideally with expertise of the analysis of peptides by liquid chromatography tandem mass spectrometry (LC-MS/MS). The post is located at the School of Life Sciences with access to a variety of facilities and its state-of-the-art mass spectrometry facilities. The project aims to identify and develop novel biomarkers for Motor Neuron Disease and related neurodegenerative diseases by exploiting deeply curated patient cohorts

from Anne Rowling Regenerative Neurology Clinic (Edinburgh) and quantitative proteomics by apply state-of-the-art Mass Spectrometry technologies (Dundee). Furthermore, the project involves in collaborating with international consortiums such Global Neurodegenration Proteomics consortium (GNPC), Gates Ventures where we will contribute and access proteomics data from large cohort of patients living with neurodegenerative diseases including motor neuron disease, Alzheimer's disease, Parkinson's disease etc., to identify disease specific biomarkers. The post holder will conduct original research and produce data for high quality publications, grant proposals and dissemination to the public. The post-holder will have opportunity to work in both labs at University of Dundee and Edinburgh. This post is ideally serves as a springboard for those motivated to pursue for independent career fellowships or industry career in future. Our recent publications on this area (PMID: Nirujogi et al., Biochem J, 2021; Fan et al., Acta neuropathol, 2021, Banerjee et al., Science Advances, 2023). The post holder will closely interact with a multidisciplinary team of translational neuroscientists, MND clinicians, drug discovery experts to translate identified biomarkers to MND- SMART clinical trial. We will employ cutting edge transcriptomics and proteomics on state-of-the-art Orbitrap Astral and tims-TOF SCP mass spectrometers and proteogenomic analysis in developing multiplexed targeted PRM assays in clinical samples. This is a fixed-term appointment for 36 months.

Enthusiastic candidate should have recently completed a PhD with a strong background and interest in neurodegeneration and mass spectrometry-based proteomics. The position will be a fantastic opportunity to work alongside with multidisciplinary team employing cutting-edge technologies such as transcriptomics, quantitative proteomics and proteogenomic analysis. The candidate will be part of ongoing collaborative research projects between MRC-PPU Dundee and University of Edinburgh. Principal Duties

• Undertaking research aimed at developing a compendium of TDP-43 mediated cryptic exons/peptides in human primary macrophages and clinical samples by employing transcriptomic, quantitative proteomic and proteogenomic analysis.
• Project development and implementation by performing primary cell culture, sample preparation for mass spectrometry analysis. Development of Parallel Reaction Monitoring assays for cryptic peptides.
• Integration of transcriptomic and proteomic data to perform proteogenomic analysis and bioinformatic analysis.
• Serum/plasma sample proteomic preparation, quantitative proteomic data acquisition and data analysis.
• Working closely with other lab members and collaborators and able to present and discuss scientific work at group meetings and scientific conference and meetings with external collaborators.

The post holder will:

• PhD with an outstanding track record with clear evidence of at least one first-authored publication in an internationally recognized peer-reviewed journal

• Strong background in Quantitative proteomics using High-resolution mass spectrometry including sample preparation and database searches using MS-Fragger, DIA-NN and Skyline software suites.
• Strong enthusiasm to neurobiology and neurodegeneration
• Prior experience in developing PRM assay is advantageous but not essential.
• Experience with R or Python and able to write, modify and implement published code and packages for data analysis and visualization.

We are recruiting for an exceptional individual to join us as Postdoctoral Research Assistant within the Maniaci Group, to develop chemical probes to uncover the molecular basis of the "cleave to modify mechanism", an unprecedented and exciting new mechanism for protein regulation that the Maniaci lab has uncovered. This is a fixed-term appointment for 24 months in first instance.

The work will focus on understanding the regulation and function consequence of the "cleave-to-modify" mechanism which involves the processing of Ubiquitin-like-domain (UBL) fusion proteins to enable their further modification as a mean to regulate protein biological function. While the mechanism underpinning the conjugation and removal of Ubiquitin is now well understood, not much is known about the proteolytic processing of UBL fusion proteins, and the biology that are revealed and triggered as a result. This project aims to elucidate the enzymatic mechanism and substrate specificity of this previously undescribed process and will involve the design, synthesis and biochemical evaluation of a library of novel benchmark chemical probes.

The successful candidate will be expected to design, synthesize and characterise novel covalent and not covalent peptide like ligands. The qualified best ligands will be developed into a variety of novel bifunctional molecules, that recruit covalently, efficiently and selectively to the protease of interest for a variety of novel bifunctional molecules, that purposes, including structural studies and activity-based profiling. You will join a diverse, fast-paced and collaborative team within the Maniaci Group to contribute to multi-disciplinary project. This is a unique opportunity to gain experience in world-leading, collaborative and cutting-edge research in one of the hottest areas of modern chemical biology and biotechnology. Project Summary:

Coastal Mangroves in Eastern and Western Africa offer coastal communities much-needed ecosystem services and protection against key climate change risks, including risks to low-lying coastal-ecological systems, risks to terrestrial and ocean ecosystems, and as a result, risks to food security. Coastal communities are particularly vulnerable to climate change impacts because of (1) their location being highly exposed to sea level rise and erosion, (2) poor socio- economic conditions, and (3) their high dependence on mangrove ecosystems. Coastal Mangroves however are highly threatened both by anthropogenic and climate change pressures. The main objective of the project (CoastMan) is co-production and uptake of a Multi-Criteria Decision Support System (DSS) that provides vulnerable communities and decision-makers a knowledge-based decision-making capacity for the restoration and conservation of socio- ecological systems. An interdisciplinary and transsectoral team is established from researchers and stakeholders from across three continents (America, Europe, and Africa), in partnership with vulnerable groups to co-develop and uptake a DSS to promote mitigation and strategies that enhance the adaptive capacities of the mangrove socio-ecological systems in Eastern and Western Africa. Drawing on quantitative and qualitative data from extensive field research in coastal Tanzania and Ghana, extensive literature, and remote sensing, the project will 1) co- develop dynamic maps of drivers and risks of mangrove habitat loss, 2) support locally-led social- ecological and climate resilience through coordinated management of mangrove resources, 3) co-develop sustainable livelihood strategies for coastal communities, 4) co-design a novel multicriteria framework for mangrove restoration and conservation, and 5) co-develop a novel DSS for mangrove socio-ecological systems. We devised a co-production and dissemination strategy so that project outputs will assist in identifying mitigation strategies and enhancing adaptive capacities of the frontline coastal communities, including building their capacity needs in designing climate actions such as restoration and conservation of mangrove ecosystems. Furthermore, the project will lead to improved knowledge and capacity of partner institutions through supporting early career researchers and promoting strengthened collaboration between partners and vulnerable coastal mangrove-dependent communities.

The role:

This role will have three primary components: 1) Social science data collection and analysis including mixed methods such as surveys, interviews, focus groups, co-produced and participatory research. 2) Managing project dissemination, communication and engagement 3) Undertaking general project research tasks, including reviewing literature, assessment of impact and contributing to project meetings. Starting salary expected to be on Grade 7. If the successful candidate does not have a PhD or equivalent experience in a relevant area, appointment will be made to Training Grade 7 (£32,332 - £34,980).
